If I want to block a few clients within a collection from getting an window update patch being advertised to the collection, can I just right click on those clients and choose block from the the pop-up menu or is there a better way to do it? Thanks

Block has nothing to do with any specific advertisements or deployments. It blocks the client from communicating with the site entirely.

So is it possible to block a windows update to a PC because there are some computers in my organization that I would like to block them from receiving Windows SP3 update.

Personally, I'd make a new collection, and explicitly exclude those computers from the collection query.  Target the  deployment of the SP3 update to the new collection.

There is no way to exclude systems from an advertisement or deployment. By design, targetting in based on an entire collection. If you need a different (sub-)set of systems grouped together to target an advertisement or deployment, then the designed method is to create a new collection - just like like Sherry said above.
